Yes the notifier will show up when an update is available if you have it checked to hide the button.

The option to hide the button is just if you do not want the buttons to always show on the toolbar. A space saver so to speak.

what happens if you uncheck updates/check for updates when Firefox starts and set Alerts/check for updates every 24 hours?

No problem at all.

It will do just that. Check for updates once a day and inform you with the visual cues if any updates are available.
