I start with Windows Add/Remove Programs. However as we all know it leaves Folders (sometimes empty but oftentimes not), as well as .dll & .dat files scattered around on my NTFS C drive. Knowing that, I use Windows Explorer and manually delete any of the aforementioned items I can see or find.

If the program is more entrenched, such as my currently installed Norton Internet Security 2008 program, first I run Registry Mechanic from PC Tools because it's not quite as aggressively scary to me as CCleaner, then I'll run CCleaner to see if it picks up anything.
One thing I always appreciate about the Polls is that they include links to the programs so I get to learn something new, which is always the case on this site.

I voted Windows Add/Remove.
I'll generally do a manual search for remnants left behind when I'm done though.

Have also been 'playing around' with Revo Uninstaller a bit, after seeing it mentioned here. (I've actually had that on a Thumb Drive for quite a while and forgot all about it...nice GUI).
