There are a slew of variables which will affect your data transfer speed from other parts of the Internet. WRT speed test sites, some of the variables are limitations inherent in the JS, or Flash implementations of the tests.
In general the most accurate speed test is the download speed reported by the client for the current file you are downloading from any given source.
