Not a CUCM expert by any stretch, but...
a) are you configuring CUE from CLI or GUI?
b) are you specifying SMTP via hostname or IP address?
c) can the CUE and the router resolve the hostname / IP address properly?
d) does the CUE and router have basic layers 1 - 3 connectivity to the SMTP server?
e) from the router, issue a "telnet 25," does it show OPEN?
Start with layers 1 - 3 connectivity, work your way from there.

From the SMTP server, can you ping the ISM &/or the CUE unit? |

Yes its reachable from SMTP server. |

Since you said telnet from the router works, do an extended telnet to the SMTP server sourcing it from the ISM interface on the router
telnet x.x.x.x 25 /source-interface ism 0/0 |

Sorry for late reply.
i have applied this command and output is: Trying x.x.x.x 25 ... Open 220 domain name.com Aug 2013 10:23:16
500 5.3.3 Unrecognized command 500 5.3.3 Unrecognized command 500 5.3.3 Unrecognized command 500 5.3.3 Unrecognized command 500 5.3.3 Unrecognized command 500 5.3.3 Unrecognized command 421.4.7.0 too many errors on this connection, closing terminal connection
now what you say... |

The message '500 5.3.3 Unrecognized command' is actually coming from the exchange server. On the exchange turn on protocol-level logging for the connector that's generating the 500 error. Then refer to the logs for further details of the session failure. Get with your exchange admin to see why the system is spitting out the 500 message. From the Cisco side you may also want to open a TAC case. |

Agree with ladino -- basically at this point what you've done is proven layers 1-4 connectivity between the ISM and the exchange server. Question is what the Exchange server is doing with the request from the ISM. Regards |
