You don't get it cause the South Park guys, as with most current comedians, totally miss the point of the joke in that they think the only point is to be as crude as you possibly can. Thus you usually get a version of the joke that pretty much any first grader can tell and be just as funny.
At least the South Park version is "honest" cause that's the whole point of the show and it is being told by little kids. On second thought maybe they do get it and this is a poke at those who don't; like what they did when George Clooney offered to do a voice.

It's funny because of it's crudeness, but other than that, I really don't get the point of the joke. It really isn't anything special. Actually, it's extremely non-special, I didn't laugh once.
